Quality control (QC) of methods and systems is necessary toward the generation of reliable mass spectrometry (MS) data. To help obtain accurate and precise metabolomic data that can be reproduced by independent laboratories with different MS technologies, standardized protocols and reagents are ultimately necessary. To that end, we are pleased to offer a metabolomics QC kit.

The metabolomics QC kit – MSK-QC-KIT – is comprised of two vials of 14 stable isotope-labeled standards.  Procedurally, these vials are to be mixed prior to use as workflow QC standards in matrix-free and matrix-containing (e.g., human plasma) analysis.  Such evaluations would involve the method (e.g., implemented at the beginning of a workflow, before and after extraction) and analytical platform, with metric tracking over time for performance accreditation. The potential use of this kit though extends beyond QC testing. It can also be used as spike-in standards for quantitation. In quantitation, an aliquot of the stock mix (diluted as necessary) could be spiked into a sample matrix for use in isotope dilution mass spectrometry (IDMS) applications.

What is the analyte composition of MSK-QC-KIT?

This kit comprises 2 vials of lyophilized metabolites mixes. Vial one contains 5 amino acids while vial two comprises a collection of 9 other metabolites (including caffeine, fatty acids, and organic acids).  Reconstituting each vial, independently, in 1 mL of solvent (e.g., 0.1% FA and 5% ACN) yields the concentrations noted in this QC kit flyer.

How many runs does the kit allow?

Possible is 100-200 runs if 5-10 μL is used per sample analysis.

Can you show representative XICs for example metabolites from LC-MS analysis of MSK-QC-KIT?

Representative XICs of a subset of kit metabolites - vial 1 in a) and vial 2 in b) - measured in human plasma by RP-LC-MS (negative ESI, Q Exactive).  Note that the stable isotope-labeled metabolites co-eluted with their endogenous analytes in the plasma analyses and their m/z were [M-H]- (as specified in isoMetlin), unless otherwise indicated in the figure.

How often should QC measurements be performed?

The QCs could be conducted 10 times prior to sample analysis to establish baseline performance and intervention limits.  During the experimental analysis, the mix could be injected after 5-10 samples to monitor the precision of retention time and intensity, for example, over the course of a 96-well plate.

If I don’t see my desired metabolites in the MSK-QC-KIT. Can a custom mix or an add-on vial be formulated?
